Enhancement of fatty acid synthesis by isoflavones in rainbow trout

Eicosapentaenoic (EPA) und Docosahexaenoic acid (DHA) are important highly unsaturated and longchain fatty acids (LCPUFA) for human nutrition due to their potential healthily effects. The LCPUFA content and their respective shares vary among different fish species and are dependent on the dietary lipid content and fatty acid composition. As rainbow trout belong to the group of freshwater species, the capability of the ALA to LCPUFA conversion is increased (in comparison to marine species). However, total deposition of LCPUFA in body tissues is lower. Thus, supplementation of these fatty acids is required to enhance product quality. So far this is done by inclusion of marine fish oil into diets. As fish oil is running short on the world market while aquaculture production is predicted to increase, sufficient supply of population with LCPUFA seems questionable. The development of alternative feed ingredients as well as inquiry and identification of the LCPUFA synthesis pathways are of the utmost importance. Novel feed ingredients, such as specific micro-algae, seem to be able to meet the requirements of LCPUFA. At the moment, production is only feasible in small amounts and involves high costs. Therefore, enhancement of endogenous LCPUFA synthesis in fish seems reasonable to reduce usage of marine fish oil and simultaneously improve supply by LCPUFA via fish in human nutrition. The aim of this project is to enhance the endogenous synthesis of LCPUFAs in rainbow trout (Oncorhynchus mykiss). Especially the contents of EPA and DHA should be increased by supplementation of isoflavones. Thereby, changes of the physiological parameteres of this species should be investigated. Furthermore, molecular mechanisms leading to an increase of LCPUFA-synthesis by supplementation of different isovflavones and their concentrations shall be elucidated.
